Biodiversity is fundamental to the stability and resilience of ecosystems; however, infectious diseases have emerged as a growing threat contributing to global species decline. In Taiwan, the endemic, Yellow-margined Box Turtle (Cuora flavomarginata) and Yellow Pond Turtle (Mauremys mutica) are increasingly impacted by habitat degradation, illegal wildlife trade, and biological invasions. Invasive species may act as reservoirs or vectors of pathogens, thereby posing health risks to native fauna and disrupting ecological equilibrium. Despite these concerns, research on infectious diseases affecting these native turtle species remains limited. This study aims to assess the prevalence and potential transmission pathways of herpesvirus, adenovirus, and ranavirus in wild and captive individuals through non-invasive sampling (oral and cloacal swabs). Molecular diagnostics using qPCR and PCR, followed by genetic sequencing, will be employed to identify pathogen strains and trace their likely origins, with a focus on evaluating the involvement of invasive species in cross-species transmission. The results will provide critical insights into infection dynamics, inform health surveillance strategies, and contribute to evidence-based conservation planning. Additionally, the study emphasizes science communication and stakeholder engagement to enhance public awareness, promote collaborative conservation efforts, and support the long-term sustainability of native chelonian populations and their habitats.
本研究與食蛇龜(Cuora flavomarginata)及柴棺龜(Mauremys mutica)的生態研究及收容單位合作,進行樣本採集與病毒感染分析。研究對象分為:(1) 原生棲地個體、(2) 野放個體、(3) 重複捕捉個體,以比較不同族群間的病毒感染狀況。
樣本採集以喉頭拭子為主,以獲取高品質病原核酸。若發現新鮮死亡個體,將解剖評估死因。所有樣本將置於 DNA/RNA Shield™ 保存液中,以確保核酸穩定,並降低病原體傳播風險。
實驗分析:本研究將從個體感染到族群傳播模式進行分析,結合親緣與GIS分析,提供疾病監測與防控策略的科學依據。
